CP-SUB with 150 ml scaffold regulating biomass via sequential bleeding

CellTank-35 is a novel packed-bed (CerCore) Continuous-Processing-Single-Use-Bioreactor (CP-SUB) designed for steady state continuous cultivation of high density adherent as well as suspension type of mammalian cells for expression or proliferation of biologics. CellTank designed ready-to-use, pre-assembled and facilitates integrated Single-Use-Sensor's and Single-Use-Pump all in a pre-sterilized and complete package. Designed for continues expression of product or proliferation of biomass by sequential product harvest or cell harvest performed by the integrated diaphragm pump. 

CellTank-35 is in the design process integrating one CerPhragm build-in Air-Operated-Diaphragm-Positive-Displacement (AODPD) driven by the Eirene software in the Clotho Drive Unit. CellTank general cultivation parameters controlled by Process-Control-System (PCS) from Cronus-PCS.

CellTank-35 general specification:

  • PET scaffold, matrix harbouring either suspension or adherent mammalian cells 
  • Polyamide cover integrated 100% fluid tight with vessel
  • Polyamide reactor body integrating two envelopes filled with polyester fibre matrix for enhanced bio-compatibility cultivating either adherent or suspension cells
  • Build in high performance diaphragm pump facilitating advanced operation such as; inoculation, media re-circulation, cell bleeding, harvest function
  • Efficient elimination of bubble to cell contact and kLa values above 40 sparging with air
  • Efficient head space to liquid surface contact for efficient CO2 control
  • Delivered high precision E-beam irradiated in dual polyester foil bags

CellTank-35 facilitate one Melpomene Air-Operated-Diaphragm-Positive-Displacement (AODPD) Single-Use-Pump (SUP):

  1. One integrated diaphragm SUP for cell inoculation, cell distribution inside the scaffold, harvest of product, harvest of cells for biomass control, harvest of cells as product

CellTank-35 facilitates the use of PG 13.5 x 120 mm classical format Single-Use-Sensor's (SUS):

  1. One pre-installed classical signal OneFerm pH sensor from Hamilton - included
  2. One pre-installed non-invasive VisiWell for dO2 Re-Usable-Sensor from Hamilton (VisiFerm not included)
  3. One port pre-installed with bio-mass sensor – in development
  4. One port for conductivity SUS
  5. One port for CO2 SUS – in development
  6. Extra port for optional pre-installed Glucose sensor 

CellTank-35 further integrate sensor's for measuring:

  • Temperature via PCS sensor in non-invasive well
  • Internal media re-circulation mass flow rotameter – SUS – manual read-out of 0.4 - 2.4 litre/min
  • Matrix pressure drop – SUS – 0 – 1,000 mm WC
  • Media level with an insulated SS electrode
